Just look on the Internet for places to see, everybodies taste is different. You should love it though, I've visited every country in Europe and Italy is my favourite for a holiday.

Colosseum is excellent as are the Spanish steps and trevi fountain
Food is amazing but can be privy stay off main streets like via nationale and try side streets when looking for somewhere to eat
Also the traffic is mental!
Where are the coed Roman baths at?

Public transport is fine but as Ace says much better to get your walking boots on and explore the city
If you're doing the Colosseum (which you have to really), you have to start at Capitoline Hill and explore the Forum and Palatine Hill on the way.
Firstly, the views over the Forum from behind Capitoline Hill are breathtaking. Secondly, for me, the best way to get a real grasp of the scale of the power and wealth of the elite through the glory days of the Empire is to walk among the remains of the palaces on Palatine Hill. Finally, I would argue that the Forum itself is more of a spectacle than the Colosseum.
It's there that you get to see the development of the Empire through the centuries, with grand churches built on and into the ruins of temples dedicated to the ancient gods. It's epic mate, honestly.
And this goes for anywhere in the world you visit, but avoid any restaurant with a Menu Turistico like the fackin plague!
Markets are generally a very good place to eat in Italy.
When you get there say 'bobbidy boobidy' to the locals.
Ace's advice is sound as well (believe it or not...)
Take the bit about the Vatican seriously if you're planning on going. It's well worth the time if you have it, because the art collections in there are the best within the borders of Italy barring the Uffizzi in Florence, but if you don't plan in advance or get there seriously early, you could easily - and I'm not exaggerating - lose half a day queuing just to get in.

this place is very authentic and highly regarded - wasn't too pricy when i went either - but it is a bit out of the centre of rome, i've only ever been driven there:

Did mine in the Sistine Chapel, we got there early and near enough had the place to ourselves so I thought fack it. Had the ring in my pocket waiting for the right moment.

ha, i think i did mine spontaneously over a plate of veal tongue with parsley sauce, so you probably win this one.
The Vatican is the best thing there. Go to the top of it, the view of Rome is unreal. As it is from the top of Castel Saint'Angelo.

Nah buy a ticket for the Vatican museum with a queue skip outside the basilica and you can see it all the same day. About €15 I think.
